Down expression of collagen VI regulated by the over expressed miRNA in bladder cancer

Collagen VI, a major extracellular matrix protein, is known to be associated with tumor growth and progression. It is highly expressed in various cancer types, however, that in bladder cancer (BC) exerted a diverse levels. In our previous study, the micro array test indicated that collagen VI alpha 1 (COL6A1) and alpha 2 (COL6A2) were down-regulated in BC. To reconfirm this event, the present study used polymerase reaction chain test to evaluate the expression levels of COL6A1, COL6A2 and their interacted miRNAs (miR-6124 and miR-4651) in BC and adjacent normal tissues. Compared to the adjacent normal tissues, mRNA expression of COL6A1 and COL6A2 was significantly down-regulated, while an obviously high expression of miR-6124 and miR-4651 was exerted in BC tissues (P<0.001, respectively). The Spearman correlation test revealed negative relationships between COL6A1 and miR-6124, COL6A2 and miR-4651 (Rho=-0.423 and -0.312, in each). Unlike other cancers, COL6A1, 2 were down expressed in BC, this is probably due to both collagen VI were regulated by the targeting miRNAs. The promising findings increase our knowledge of cancer biology, provide novel diagnostic and therapeutic interventions for BC.
